Tough call on this. Most of the one's I think of involve a series of plays like Bama's block of a UT field goal followed by Philip Doyle's field goal to win the game 9-6 in 1990. https://www.youtube.com/watch?v=f32wHHqwMco

We're fortunate as Alabama fans to have many great moments to relish in over the years. I wasn't yet born when "The run in the mud" occurred. The first such moment I remember was "The goal line stand". Then there was "The kick" and "wrong way bo". Sprinkled in were great moments like 315 and 323 (still the hardest hitting game I can ever remember watching).
Of course, not all these are necessarily "great plays" but all are great moments.
Teague running down LT was the single greatest play I can remember and it happened in the single greatest game I can remember.
Shaun Alexander had a great catch and run in the '98 Iron Bowl. I was there and thoroughly enjoyed it.
Shula to Al Bell (Georgia and Ohio St).
1987 2 point conversion vs Penn St. Incredible play.
2010 vs TX, Marcell Darius interception and Eric Anders sack. The play that won the Iron Bowl that year was also great.
2007 vs Arkansas we score a TD to win the game. We all knew we would be back after that (we knew beforehand, but that's when we began to see results).
Julio TD against LSU in 2008.
Ingram on screen pass vs Fla 2009.
Richardson TD run vs LSU in BSCNCG. Finally! A TD!
All the others posted are great as well. And we've just scratched the surface.

One of my all time favorite plays is one that will probably never be shown in a highlight reel because of how the game ended. Had we won the 2013 iron bowl the highlight of that game would have been the 99 yard TD pass from McCarron to Cooper. Not only was it as long of a play as you can possibly run, but it was a masterful call in a stressful situation. AJ slightly underthrew it, but it was still a thing of beauty. It was a testament to the concentration, hands, balance, and speed of Coop.

Sadly it will forever be largely forgotten because of the one play that will remain unmentioned. I remember coming unglued when it happened. I pretty much ran around my living room in excitement, thinking that was the play that would crush the barners. Boy was I dumb to assume that our talent could beat their pact with the devil. Another one I'll throw out there didn't mean much in the grand scheme of things, but daggum it was fun to watch. The Richardson juke was brought up earlier, and it was one of the prettiest moves I've ever seen at any level. But Trent had another run against Arkansas that was just ridiculous. He got hit by pretty much the entire Arkansas D, (I'm no Selma but if I remember correctly I think it was in the neighborhood of 8 defenders) and he still took it to the house.

I don't think anyone mentioned "The Kick" by Van Tiffin. It wasn't the greatest "play" in Alabama history but perhaps the greatest finish. It took some clutch plays to get in position for the kick. As Keith Jackson said when it was over "They don't get any better than this one". Here's the final drive:
